Why does genomic imprinting silence one parental copy so only the other is expressed, and how is that silencing maintained?
Imprinted genes acquire parent-specific DNA methylation marks during sperm or egg formation, and after fertilization the two alleles carry different epigenetic states. Silencing is usually enforced when a differentially methylated region recruits CTCF or Polycomb complexes, blocking enhancer-promoter communication. During cell division DNMT1 copies the methylation pattern onto the new strand, keeping the imprint stable across the lifespan.

How does a female mammal silence one entire X chromosome in each of her cells, and why does she end up a mosaic?
Early embryos count their X chromosomes and randomly select one per cell for inactivation, condensing it into the Barr body. The long noncoding RNA Xist coats the chosen chromosome in cis, recruiting Polycomb complexes that deposit repressive H3K27me3 marks followed by DNA methylation at promoter CpG islands. Because descendants inherit the silenced state clonally, neighboring tissues can express different alleles, famously visible as calico cat coat patterns.

Why do mouse embryos need genes from both parents? Genomic imprinting reveals parental tug-of-war.
Most genes treat both parental copies equally, but about one percent carry parent-of-origin marks (imprints) expressing only one side — unique to mammals. The evolutionary reading is parental tug-of-war: paternal genes push embryos to extract maximum maternal resources (his genes may ride other offspring), maternal genes balance this litter against future ones. The evidence is stark: mouse embryos with only paternal genomes grow huge placentas but stall, maternally-only ones build big embryos on skimpy placentas — two genomes negotiating, neither optional. Imprint errors cause syndromes like Beckwith-Wiedemann and much of cloning difficulty.

How do epigenetic modifications like DNA methylation affect gene expression without changing the sequence?
DNA methylation typically adds a methyl group to cytosine in CpG islands, recruiting repressive proteins and blocking transcription-factor binding to silence genes; such marks are stably inherited through cell division and shaped by development and environment without altering the base sequence.

How do chromatin-remodeling complexes regulate gene accessibility?
ATP-dependent remodeling complexes slide, evict or restructure nucleosomes to alter how DNA is wrapped around histones, making promoters more or less accessible to transcription factors and thereby regulating transcription without sequence change.

What is epigenetics, and how can a person's experience pass to offspring without changing DNA?
The DNA sequence is the letters; epigenetics are the “annotations”—methylation and histone marks switch genes on/off without altering letters. Famine or stress can shift these marks and possibly persist a generation or two, yet rarely rewrite the code permanently.

Why can two cells carrying identical DNA sequences show vastly different levels of gene activity?
DNA methylation adds methyl groups to cytosines, mostly at CpG dinucleotides, and dense methylation in a promoter generally suppresses that gene's transcription. Because the underlying base sequence stays untouched while gene output changes, the mark qualifies as epigenetic. DNMT enzymes install the pattern, and maintenance methylation copies it onto daughter strands after each round of DNA replication.

Identical twins share almost the same DNA — why do their personalities and health diverge with age?
Epigenetics supplies a key part of the answer: beyond the DNA sequence, methylation marks and histone modifications decide which genes are on or off, and those marks keep changing with life experience. Comparing identical twins, researchers found the epigenetic patterns nearly overlap in the young but diverge across hundreds of genes in older pairs — diet, stress, smoking and exercise all leave traces. Identical genes mean an identical score; epigenetic marks act as the conductor adjusting every performance, so twin lives diverge.
