How does CRISPR-Cas9 technology enable precise gene editing?

CRISPR-Cas9 uses a guide RNA to recognize a target DNA sequence and directs the Cas9 enzyme to cut the DNA there, allowing scientists to knock out or replace specific genes. Derived from a bacterial antiviral immune mechanism, it was elucidated in 2012 by Doudna and Charpentier, who won the 2020 Nobel Prize in Chemistry.

How does PCR amplify a trace of DNA millions of times?

The polymerase chain reaction uses primers flanking the target and cycles through denature, anneal and extend, so the polymerase doubles the template each round, growing exponentially. Mullis invented it in 1983 and won the 1993 Nobel Prize.

What are the roles of telomeres and telomerase?

Telomeres are repetitive DNA sequences at chromosome ends that act as protective caps, preventing degradation or erroneous fusion of chromosome tips. Telomerase is an enzyme that extends telomeres and is active in stem cells and most cancer cells; its decline is linked to cellular aging, a topic honored by the 2009 Nobel Prize in Physiology or Medicine.

What is mRNA, and how does the cell translate it into protein?

Messenger RNA copies the coding sequence from DNA to the ribosome; the ribosome reads each codon and recruits the matching transfer RNA's amino acid, extending the chain until a stop codon. Crick set out the central dogma in 1958; Watson et al. detail the process in 'Molecular Biology of the Gene'.

What is alternative splicing?

A gene's pre-mRNA can be spliced in different ways to yield several mature mRNAs and proteins, letting fewer genes create more complexity. Berget, Moore and Sharp discovered this in 1977; Lodish details its regulation.
