How is an action potential generated and conducted?
When a neuron is sufficiently stimulated, voltage-gated sodium channels open and Na+ influx rapidly depolarizes the membrane; then potassium channels open and K+ efflux repolarizes it, producing a spike. The action potential travels along the axon without decay, and its all-or-none nature ensures reliable signaling.

Why do midbrain dopamine neurons fire for reward prediction error rather than reward itself, and how does that serve as a teaching signal for learning?
Dopamine firing scales with actual reward minus expectation, not with pleasure itself. Recording in the substantia nigra pars compacta and ventral tegmental area of alert monkeys, Schultz saw bursts to unpredicted juice; after training, activity shifted to the predictive cue and vanished when reward arrived as expected, while omitted rewards drove firing below baseline. These three patterns implement the Rescorla-Wagner rule, letting dopamine act as a teaching signal that drives synaptic plasticity and policy updating.

Why does the myelin sheath wrapped around an axon greatly speed up nerve impulse conduction?
Myelin, wound around axons by Schwann cells or oligodendrocytes, is a fatty, low-capacitance insulator that sharply reduces transmembrane current leakage. Action potentials are re-triggered only at the bare nodes of Ranvier between myelin segments, where voltage-gated sodium channels cluster, so local currents leap rapidly down the axon interior to the next node. This saltatory conduction raises speed tens of times over and conserves the energy spent rebuilding ion gradients. The conduction defects of multiple sclerosis stem from damaged myelin.

When a nerve impulse reaches a synaptic terminal, how does the signal cross the roughly 20-nanometer gap to the next cell?
The arriving impulse depolarizes the terminal, opening voltage-gated calcium channels so that calcium influx acts as the trigger. With help from calcium sensors, SNARE complexes on vesicles dock and fuse the synaptic vesicle with the presynaptic membrane, releasing neurotransmitter by exocytosis; the transmitter diffuses across the cleft and binds postsynaptic receptors, opening ion channels that generate graded potentials. Because only the presynaptic terminal releases transmitter, transmission is inherently one-way and carries a delay of about half a millisecond—the universal mechanism of communication in the nervous system.

Why is synaptic transmission quantal, and how does a nerve terminal release acetylcholine one synaptic vesicle at a time?
Because transmitter is packaged in synaptic vesicles and released whole. At the frog neuromuscular junction Katz and colleagues recorded spontaneous miniature end-plate potentials whose amplitudes came in integer multiples of a smallest unit, showing that one vesicle dumps several thousand acetylcholine molecules at once. When an impulse invades the terminal, calcium channels open, calcium binds synaptotagmin to loosen the SNARE clamp, and vesicles fuse with the presynaptic membrane at active zones in an all-or-none exocytotic event.
