Why can diffusion models design never-before-seen functional proteins from scratch, and where does RFdiffusion outperform earlier approaches?
It learns the breaking and rebuilding of structures as prior knowledge. Watson and colleagues turned an image diffusion model into a protein-design engine: during training, noise corrupts real backbone coordinates step by step and the network learns to reverse each step; at generation time it starts from pure noise and denoises brand-new backbones, whose foldability is then screened with RoseTTAFold-class predictors. Compared with Rosetta pipelines built on hand rules and fragment assembly, it generates on demand under conditions such as symmetry or specified binding motifs, lifting experimental success rates by roughly an order of magnitude.

How did scientists engineer brewer's yeast into a cell factory that produces artemisinic acid, the precursor of antimalarial artemisinin?
By importing Artemisia pathway genes and rewiring yeast metabolism. Keasling's team moved amorphadiene synthase plus cytochrome P450 oxidation genes from Artemisia annua into Saccharomyces cerevisiae, up-regulated rate-limiting HMG-CoA reductase, and shut down the competing sterol branch ERG9, so carbon flux pours into the sesquiterpene route and fermenters accumulate artemisinic acid, which simple hydrogenation converts to artemisinin - a founding example of design-build-test synthetic biology.

Why is JCVI-syn1.0 from the J. Craig Venter Institute hailed as the first cell controlled by a chemically synthesized genome, and how was it built?
By rebooting a recipient cell with wholly synthesized DNA. After sequencing the Mycoplasma mycoides genome, researchers chemically made it in segments, stitched them into one circular chromosome through yeast homologous recombination, and transplanted it into an emptied shell of Mycoplasma capricolum. Early failures were traced to a single deleted base in one fragment; once repaired, the synthetic genome took over metabolism and division, and cells multiplied purely on the artificial blueprint - proof that sequence alone can carry life's instructions.

How does the genetic toggle switch that Gardner and Collins built in Escherichia coli work, and what did it mean for synthetic biology?
Two mutually repressing operons form a bistable gene network. LacI shuts down tetR while TetR reciprocally switches off lacI, so whichever repressor dominates locks the cell into that state heritably; adding IPTG or anhydrotetracycline flips the switch to the other stable state, with cooperative repression conferring flip-flop-like hysteresis and memory. This work first showed that regulatory circuits can be rationally designed like electronic ones, laying the engineering foundation of synthetic biology alongside the same-year repressilator.
