Influenza reappears with a fresh face every few decades while vaccines need annual reformulation—what two very different scales of evolutionary event lie behind antigenic drift versus antigenic shift?
Antigenic drift is the year-on-year accretion of point mutations in the hemagglutinin and neuraminidase genes, with immune pressure selecting escape variants—a gradual, incremental process. Antigenic shift instead rides on cross-species reassortment of the eight-segmented genome: when human and avian strains co-infect one cell, random segment packaging can deliver, in a single step, a recombinant wearing wholly novel surface antigens. Drift accounts for seasonal epidemics, shift for sporadic pandemics, and together they force periodic vaccine renewal.

Why do RNA viruses circulating in an infected host form a shifting cloud of competing yet complementary mutants rather than a population of identical particles?
Because their RNA-dependent RNA polymerase lacks the proofreading exonuclease found in DNA polymerases, each replication cycle introduces mismatches at roughly 10^-4 to 10^-5 per nucleotide. This high error rate seeds every genome with new mutations, so the population behaves as a quasispecies—a dynamic distribution centered on a master sequence plus its close variants. Such diversity lets the virus respond rapidly to selection imposed by antibodies or antiviral drugs.

Why do RNA viruses mutate far more rapidly than DNA viruses, and is this high mutation rate purely advantageous to the virus?
RNA-dependent RNA polymerases lack the 3-prime-to-5-prime proofreading activity of DNA polymerases, and RNA is chemically less stable than DNA, so RNA viruses mutate at rates orders of magnitude higher per site. The resulting mutant swarm, or quasispecies, lets these viruses quickly evade host immunity and antiviral drugs. Yet the mutation rate has an upper bound: past the error threshold, genomic information collapses under accumulated mistakes—which is exactly how mutagenic drugs such as ribavirin push hepatitis C virus into lethal mutagenesis.
